I don't recommend manually installing graphics. It's kind of a pain, and if you install a graphics pack that's made for an older or newer version of Dwarf Fortress than you are using, you could end up with a corrupt game and corrupt save files. With Lazy Newb Packs you are getting a bunch of graphics packs that should be compatible with the included version of Dwarf Fortress. And it makes the installation and switching of graphics packs super easy.
If you need Dwarf Therapist and DFHack, you will want a pack for v0.43.03. If you don't need those utilities and want the latest version of Dwarf Fortress, then you want a pack for v0.43.05.

If you really want to install your own graphics, you basically just copy the files from the graphics pack into the corresponding locations of the Dwarf Fortress folder.
- Delete your /Dwarf Fortress/raw/graphics folder
- Drag the /raw/graphics/ folder from the graphics pack into your /Dwarf Fortress/raw/ folder
- Drag the files from the graphics pack's /raw/objects/ folder into your /Dwarf Fortress/raw/ folder
- Drag the files from your graphics pack's /data/art/ folder into your /Dwarf Fortress/data/art/ folder
- Drag the files from your graphics pack's /data/init/ folder into your /Dwarf Fortress/data/init/ folder
That should be everything you need to install. There may be some other files, but they are optional. Most notably, there may be some kind of dfhack.init or onload.init type of file in the /raw/ folder of your graphics pack. This probably contains some custom config settings for Text Will Be Text. If you want this copy it into your /Dwarf Fortress/raw/ folder.
Make sure the graphics pack you are installing is compatible with the specific version of Dwarf Fortress that you are using. A graphics pack that says it's for DF v0.43.05 should not be used with DF v0.43.03 unless the graphics pack author says it's safe to do so. If you really want to install a graphics pack that is not compatible, then copy over everything except for the /raw/objects/ files. The /raw/objects/ files are the ones that cause problems if they are not for the same version of Dwarf Fortress that you are using.
