Doing my Pacifist run on Deus Ex: Human Revolution. I probably won't get Foxiest of the Hounds anytime soon, though.

there was some uber challenge for Legend of Zelda: Ocarina of Time that I tried.

It was hella hard and relied on a tooooon of glitches to work. I don't think I ever finished.

No thanks.  I'm all for a challenge, but I'm not a masochist.   :)  Learning how to do the glitching necessary for this would be interesting though.

I have considered doing a 3 heart run, but I've never convinced myself to start.

Anyone considered doing a speed run of Narbacular Drop?

I think there's a 20 second speed run out there.  The record is insanely low.

Alternatively, get the only movable object in the game (a physics cube in the first room) all the way to the end.

what was it called - the pokémon thing - only catching the first pokémon you encounter per route and releasing all that were defeated ... yeah, i'm planning one of those in Black.The Game which i ordered recently.
the last self-imposed challenge was playing through divine divinity without ever equiping a weapon.
before that: Naked arcanum. I got till tarant ...

New Vegas with one life, and extremely lethal bullet damage for enemies (1 shot is all it takes from the big ones). No DR.

Skyrim with no potions, sneaking, reloading, armor rating and most of the toughest mods there are.

Baldur's Gate 2 with ALL the unfair and/or "meant for normal difficulty" mods played on Insanity, no reloads, no level 9 spells in SoA, no HLA's at all, one dice roll for character attributes, rest of the party made by other players and thus having no influence on who do you get, no cheesy items or tactics. Complete ban on things like "protection from evil" and "protection from magic". Rest only after boss fights.

Dead Space 2 on zealot in one sitting, no reloads or checkpoints, blind.

Dark Messiah Hardcore with one life.

Rome Total War... Play as the worst (unplayable) faction (vanilla or modded) that has not a single top tier unit, don't attack major factions until they pour endless stacks your way, give Roman and other Major Power cities enough pop and process_cq them for total development and 24000 pop in the first few years. No trading. War with everyone.
There was more to Rome, but it's a blur now.

Warhammer Online (Now dead) Use one hand to beat an enemy.

These would be the more memorable ones i've done/am doing.

Its called a Nuzlocke. The name is combination of Nuzleaf(The "mascot" of the first nuzlocke) and some character from Lost(the guy who came up with the idea loved Lost).

You have not known hell until you tried to get into Gerudo valley using a fuckign cucco.

Somwhere, on my N64 cartridge for OOT, there is my uber run, stopped dead in it's tracks at the entrance to the fucking gerudo valley.

Play Mount & Blade on 1:1 damage, unarmed, on a horse, alone. I dare you. I once managed to take a castle unarmed and alone! I had about 3% of my health left and had a few dozen assorted pointy things jammed in my chest.

Heres a few I have tried atleast.

X-Com superhuman Antarctica challenge.
-Difficulty set to superhuman.
-You may only have one base and it must be in the middle of Antarctica
-No reloads. Whatever happens, happens.
I failed it but I will try it again at some point when I am bored.

Icewind Dale 2 mercenary band.
-Hardest difficulty.
-Party must consist of 6 fighters.
-No savescumming, what happens, happens.
-If a party member dies, you must replace him with a new fighter character.

Baldurs Gate 2: Solo run.
I thought this would be harder than it was but after a while I breezed through it with a sorcerer.

Wizardry 8: (if you haven't played this game, do, it's great, the only downside is that it's the third in a trilogy and the previous two are DOS games)
 - Get both the T'rang and Umpani NPCs to join your party.
(This one is theoretical, last I checked, as no one skips out on the last mission from each of those races; 100k experience for each party member?  SURE, I'M MAKE ETERNAL ENEMIES OF BOTH EMPIRES!  I also haven't checked in several years)

Torin's Passage
 - Completion.
This is neigh on impossible, due to one puzzle.  Play "find the spot" with audio clues.  Not even "hot!"/"cold!" but "yay!"/"nay!"  Good luck finding the third spot (I found it by accident).  Last I checked, there was no readily available solution (i.e. screenshot and the three spots outlined).

About to revisit Final Fantasy 7.  Remember how the back of the manual had an advertisement for memory cards saying something along the lines of "Just try beating FF7 without one"?
