Infrastructure because we have no vehicles to launch yet.
Macrobiology because our talented workforce ought to be a talented workforce IN SPAAACE.
Normal space missions for now because they will be the easiest to set up while building infrastructure: something we can work on and learn from while ground research toward manned missions progresses. (I have a feeling this is less Spamkingdom and more Kerbals; we will want knowledge in all three facets to thrive.)

I think we should focus on the bread and butter of space colonization - the land bodies. Moons, worlds and asteroids, all of which is covered under  colonization. Sure we can assemble orbital systems, but I feel like the faster we push towards a colonial presence, the better we are off.
