No.
You can take over individual sites, but you can't run a civilization. Not yet anyway, possibly in a fast future update.

I wonder if it would be possible to become a leader by joining a dying civilization and killing the last few citizens or something

I wonder if retired adventurers could also become leaders.

You can. I haven't had it happen with an active adventurer, but repeatedly assassinating the Law Giver of some humans eventually led to a retired adventurer being chosen.
